You have to fool tufte-book that natbib is loaded and also
biblatex that natbib is _not_ loaded:

\@namedef{ver at natbib.sty}{9999/12/31}% declare natbib as loaded
\makeatother
\documentclass{tufte-book}
\makeatletter
\expandafter\let\csname ver at natbib.sty\endcsname\relax% reverse
\makeatother
\usepackage{biblatex}
> \addbibresource{refs.bib}

> \title{An Example Document}
> \author{Adrian F. Clark}
